Inhibin beta A chain; Activin beta-A chain; Erythroid differentiation protein
Activin A is 100% identical between human, mouse, rat, pig, cow and cat proteins.
Description: Activins are homodimers or heterodimers of the various β subunit isoforms, belonging to the TGFβ family. Mature Activin A has two 116 amino acids residues βA subunits (βA – βA).
Molecular weight: 27.4 kDa disulfide-linked homodimers of two βA chains, each containing 116 amino residues.
Figure 1. SDS-PAGE and Western Blot analysis of recombinant Activin A. Samples were loaded in 15% SDS-polyacrylamide gel and stained with Coomassie blue. Lane MWM: Molecular weight marker (kDa); Lane 1: contains 500ng of recombinant Activin A. Lane 2: Western Blot analysis with specific anti-human Activin A contains 250ng of rhuman Activin A.
Biological functions: Activin exhibits a wide range of biological activities, including mesoderm induction, neural cell differentiation, bone remodelling, hematopoiesis, reproductive physiology, cell proliferation, differentiation, wound healing, apoptosis, metabolism, etc. Activins plays a key role in the production and regulation of hormones such as FSH, LH, GnRH and ACTH. Cells known to express Activin A include fibroblasts, endothelial cells, hepatocytes, vascular smooth muscle cells, macrophages, keratinocytes, osteoclasts, bone marrow monocytes, prostatic epithelium, neurons, chondrocytes, osteoblasts, Leydig cells, Sertoli cells, and ovarian granulosa cells.
- Activin A is measured by its ability to inhibit mouse plasmacytoma cell line (MPC-11) cells proliferation. ED50 ≤ 10 ng/ml.
- Effect of rh Activin A on keratinocytes differentiation: rh Activin increases the expression of E-cadherin and improves the distribution of e-cadherin in cell-cell contact areas.
Figure 2. Expression of E-Cadherin by immunofluorescence. Untreated cells; Positive control of differentiation; rh Activin A ( 20 ng/ml). Rh Activin increases the expression of E-cadherin and improves the distribution of e-cadherin in cell-cell contact areas.
Activin A is very commun used to culture embryonic stem cells (ES cells: Neuronal Cells, Hematopoesis, Inflammation, etc) and keep them in an undifferentiated state.
It is also realted to the following diseases: Rheumatoid Arthritis, Hepatitis C, Atherosclerosis, Spinal Cord traume, Type 2 Diabetes, Irritable Bowel Syndrome
